摘要
目的探讨川芎嗪对高糖诱导的人近端肾小管上皮细胞HK-2转分化的影响。方法将HK-2在体外与高糖(25mmol/L的D-葡萄糖)、高糖+川芎嗪(40/80/160μg/ml)共培养48h后,用显微镜观察细胞形态的改变;用间接免疫荧光法观察α-平滑肌肌动蛋白(α-SMA)的表达;RT-PCR检测α-SMA和E-钙黏素mRNA的表达。结果经高糖刺激后,HK-2形态由立方形铺路石样转变为梭形长条样;加入80μg/ml川芎嗪共同干预后,细胞形态无明显改变。免疫荧光检测显示,经高糖刺激后,α-SMA表达明显增强;加入80μg/ml川芎嗪后α-SMA表达明显减弱。RT-PCR检测显示,经高糖刺激后α-SMAmRNA表达显著增加,E-钙黏素mRNA表达显著减少,与正常对照组比较差异均有统计学意义(P<0.01);高糖+不同剂量川芎嗪组HK-2中α-SMAmRNA和E-钙黏素mRNA表达水平组间两两比较,差异均有统计学意义(P<0.05)。结论川芎嗪可以阻断高糖诱导的肾小管上皮细胞转分化,进而抑制糖尿病肾病的肾间质纤维化,且存在剂量依赖关系。
