玉米优良自交系基因枪转化受体系统的建立 被引量:8

摘要 研究了影响玉米基因枪转化受体系统效率的若干因素 ,建立了玉米优良自交系的稳定、高效胚性愈伤受体系统 ,获得了正常结实的转基因植株 .在培养基、激素成分配比适当 ,较低温继代 ,有利于愈伤胚性化 .讨论了筛选压的确定方法和原则 ,发现梯度施加筛选有助于提高再生转化频率 .对转基因植株进行了PCR分子检测 ,证明目的基因已整合进玉米基因组中 . Several factors affecting transformation of elite maize inbred lines were studied, and an efficient and stable transformation system of embryogenic calli was established. Transgenic calli and regenerants were selected on medium with Hygromycin in a step wise manner. Several fertile transgenic plants were obtained. The results of PCR certified the integration of target gene into maize genome.
