荧光原位杂交在神经母细胞瘤中的应用

摘要 目的 :检测神经母细胞瘤 ( NB) 1号染色体短臂杂合性缺失 ( 1p L OH) ,并观察 1p L OH与临床预后的关系。方法 :应用荧光原位杂交技术 ( FISH)——用非放射性物质标记核酸探针 ,根据核酸杂交原理 ,在间期核和染色体上检测特异性 DNA序列。结果 :检测 2 0例神经母细胞瘤患者的 1p L OH中 6例有 1p L OH,占 30 % ,并观察到1p L OH大多发生在年龄大于 2岁的 期 / 期的神经母细胞瘤患者中。结论 :神经母细胞瘤在 1p36 .2~ 36 .3区域内有杂合性缺失 ;1p L OH与 NB分期密切相关 ( P <0 .0 1) ;1p L OH与病人确诊时的年龄密切相关 ( P <0 .0 5 ) ;1p L OH与 NB的预后密切相关 ( P <0 .0 5 )。 Objective:To detect lp LOH in neuroblastoma and study the relation of 1p LOH and prognosis of NB. Methods:The authors detected 1p LOH in 20 cases of NB by fluorscene in situ hybridization. FISH is a technology which can detect specific DNA sequence in chromosome and meta nucleus. Results:1p LOH was found in 6 of 20 cases (30%). 1p LOH is often found in more than 2 year old and Ⅲ/Ⅳ stage NB patients.
