反义核酸技术在胶质瘤治疗中的进展

摘要   反义核酸(Antisense nucleic acids)技术是分子遗传学和肿瘤基因治疗上颇受重视的课题.遗传学者以反向遗传学角度,利用反义核酸特异性阻断细胞内单一基因表达,研究特定基因的功能,不影响其它基因的正常功能.因而,此技术对阐明单个基因在肿瘤发生、发展中的作用有重要意义;反义核酸技术也是基因治疗中较具吸引力的手段之一.利用此项技术 ,针对与肿瘤发生、发展起重要作用的几种癌基因或抑癌基因的突变体以及自分泌生长因子及其受体的基因,设计反义核酸,在转录或翻译水平封闭异常基因,使其编码产物不表达或低表达,阻断肿瘤细胞内异常信号传导,使肿瘤细胞转入正常轨道或抑制肿瘤细胞生长或诱导肿瘤细胞凋亡.  ……
