双特异性单抗异聚体促进红细胞清除循环致病原 被引量:3

摘要 论述了灵长类红细胞抗 - CR1单抗 (抗 - CR1m Ab)与抗致病原单抗 (抗细菌或病毒 m Ab) ,经过硫醚或生物素基化并经抗生蛋白链菌素 (SA)连接 ,形成双特异性单抗异聚体 (HP)。此 HP不但与灵长类红细胞上成簇分布的 CR1分子特异性结合 ,同时也与致病原单抗特异性地结合 ,从而使红细胞与致病原间接结合形成复合体 (E- HP- Ag)。此复合体一旦形成 ,绝大部分循环中的致病原 (>90 % )可被迅速清除 ,在肝脏中迅速销毁 ,同时伴有 ECR1的永久性损失 ,显示出红细胞强大的免疫调理功能。在国外已形成目前治疗血源感染性疾病 (特别是缺乏有效药物治疗的病毒性感染疾病 )最有前途的研究领域之一。
