基于SPR原理的生物分子相互作用光强法检测技术

摘要 本文简述了表面等离子体共振(Surface Plasmon Resonance,简称SPR)原理、激发表面等离子体共振的条件和共振发生时反射光的光强变化,对角度扫描法和波长扫描法进行了分析。接着,基于角度扫描的光强检测,以半导体激光器为光源,建立了生物分子相互作用检测实验装置,并在模拟分析的基础上以蓖麻毒素与其单克隆抗体为样本进行了实验。实验表明,实验系统结构简单,可以实时监测生物分子间的相互作用过程,对样本无需进行标记,操作简便。 The principle of SPR(Surface Plasmon Resonance),the excitation condition of SPR phenomenon and the phase variation of the reflected light in SPR are presented. Angle scanning and wave length scanning are analyzed and compared. An equipment with semi-conductor laser based on angle-scanning is set up to detect the intensity of the reflected light. Based on a simulation analysis of the biomolecular interaction, different expriments are completed on the interaction of ricin and its antibody. The experiments demonstrate that the equipment is simple, easy to operate and suitable for probing biomolecular interaction in real time and in site without the requirement on labelled molecule.
