大鼠坐骨神经损伤后NGF基因表达的变化 被引量:9

摘要 目的 探讨大鼠坐骨神经损伤后NGF基因表达的变化。方法 采用RT-PCR法,半定量分析坐骨神经横断后1d、1周、2周、4周远侧端组织中 NGF mRNA含量的变化。结果 大鼠正常坐骨神经中存在 NGF mRNA.但表达量较低。坐骨神经横断后.其远侧端组织中 NGF mRNA表达量显著升高,而且呈现一种特殊的“双相增高”现象。结论 坐骨神经损伤后,NGF基因的表达呈现出双相增高现象。 Objective To evaluate the expressive variety of NGF gene in injured sciatic nerves of rat.Methods For hemi-quanti-tative anal ysis,RT -PCR method was used in the detection of the levels of NGF mRNA in the distal s tumps at 1 day and 1,2,4 weeks following transection of sciatic nerve. Results The level of NGF mRNA was low in the normal sciatic nerve.But it was increased in the distal stump after sciatic nerve transection.The biphasic i ncrease in NGF mRNA was characterized by a first very rapid and transient increa se peaking at ist day after lesion,than a second prolonged and slow eleva tion starting around ist weed after lesion.Conclusion The p rocess in NGF gene expression after sciatic nerve lesion was biphasicly incre ased.
