摘要
目的 观察实验性铅中毒对小鼠海马神经元性一氧化氮合酶(nNOS)基因表达的影响,探讨铅致学习记忆损害的分子毒理学机制。方法 采用RT-PCR方法,半定量分析正常及铅染毒2、4、8周小鼠海马nNOS 基因表达的变化。结果 nNOS mRNA存在于正常小鼠海马组织中;铅染毒后 2周,海马 nNOS mRNA的含量显著减少;随着染毒时间的增加,海马 nNOS mRNA的含量进一步逐渐下降。结论 铅可影响海马 nNOS基因的表达,其表达量随染毒时间的增加而呈指数下降趋势。
Objective To study the effects of lead exposure on expression of nNOS gene in the mouse hippocampus,and to investigate the molec ular toxicological mechanism affected on the learning and remember by lead expos ure .Methods Using the reverse transcription-polymerase chain rea ction (RT-PCR)metho d,as a hamipuantity assay,nNOS mRNA levels in the hippocampus of normal and lead e xposure two,four,eight weeks mice were investigated.Results nN OS mRNA presented in the normal mouse hippocampus,nNOS mRNA levels in the hippocampus were markedl y d ecreased by lead exposure two weeks and gradually further decreased with lead ex posure time increasing.Conclusion Lead can affect the expression of nNOS gene in the mouse hippocampus with a decrease in a index way following the time of lead exposure.
