切应力作用下联合培养的血管平滑肌细胞对内皮细胞分泌t-PA和PAI-1的影响 被引量:3

摘要 目的 探讨切应力作用下联合培养的血管平滑肌细胞对内皮细胞分泌t-PA和 PAI-1的影响。方法 应用发色底物法研究 40dyn/cm2切应力作用下与血管平滑肌细胞联合培养的内皮细胞分泌 t-PA和 PAI-l的量的变化,以静态条件下单独培养的内皮细胞和联合培养的内皮细胞为两对照组。结果 静态条件下联合培养的内皮细胞分泌t-PA的量较单独培养的内皮细胞分泌的量显著增多.同期PAI-l的分泌量减少。切应力作用下联合培养的内皮细胞t-PA的分泌量进一步增多,PAI-l的分泌量在整个切应力作用过程中没有显著的变化。结论 切应力作用下,与血管平滑肌细胞联合培养的内皮细胞的抗血栓能力增强。 Objective The effect of vascular smooth muscle cells (VSMCs)on the abilities of secreting t-PA and PAI-1 by Co-cultured endotheli al cells(ECs) under shear stress has been studied.Methods Contents o f tissue plasminogen activator (t-PA)and type l plasminogen activator inhibitor ( PAI-1)by Co-cultured ECs with VSMCs under shear stress were investigated with th e methods of colorsubstrate,two controls of ECs cultured alone and Co-cultured E Cs were maintained under static conditions.Results Compared with ECs cultured alone,in static condition,the content of t-PA secretion by Co-cul tu red ECs increased dramatically,while at the same time,the content of PAI-1 decr ea sed.Under shear stress,the t-PA secretion by Co-cultured ECs increased further .The PAI-1 secretion was unaffected by shear stress over the entire time range. Conclusion Under shear stress condition,the capabilities of an ti-thrombosis of Co-cultured ECs were strengthened,and our results may also su pply some thoretical and experimental foundation for improving tissue engineerin g of vascular endothelial cell seeding.
