Mn9202对培养大鼠胃底平滑肌细胞的5-HT、TH及其受体的影响 被引量:1

摘要 目的 研究二氢毗陡类钙桔抗剂,[2,6 二甲基-4-(3-硝基苯基)-1,4-二氢-3,5-吡啶二羧酸-3-甲酯-5-正戊酯(Mn9202)了、拉西地平(lacidipine)对培养的大鼠胃底平滑肌 5-HT、5-HT合成酶(TH)及 5HT受体(5-HTR)的影响。方法采用免疫细胞化学和图像分析等方法观察Mn9202和lacidipine作用下培养的大鼠胃底平滑肌细胞中5-HT、TH和5-HTR含量的变化.并与5-HT的桔抗剂赛庚定(cyprohetadine)比较。结果 正常大鼠胃底平滑肌细胞5-HT、TH、5-HTR免疫反应均呈阳性,而Mn9202、lacidpine和cyprohetadine均能使胃底平滑肌5-HT、TH和5-HTR的含量减少。结论 大鼠胃底平滑肌胞具有自分泌5-HT的功能,Mn9202和拉西地平可能通过减少5-HT的合成而使5-HTR的表达下降,这一结果与5-HT桔抗剂赛庚定相似。 Objective To study the e ffects of DHPs calcium antagonist Mn9202 and lacidipine on 5-HT、TH and 5-HTR of rat stomach fundus smooth muscle cells.Methods It's effects was investigated by using immun ocytochemistry and image analysis of stomach fundus smooth muscle cells in culture and compared with cyprohetadine(antagonist of 5-HT).Results 5-HT、TH and 5-HTR immunoreactive substances were existed in SFSMC.The results of image analysis indicated that the content of 5-HT、TH and 5-HTR were significantly lower when using Mn9202、lacidipine and cyprohetadine.Conclusion The results sugg est that the rat stomach fundus smooth muscle cells possess 5-HT autocrine function,and DHPs calcium antagonist reduce the number of 5-HT receptor in the rat stomach fundus smooth muscle cells though the reduction of 5-HT sythesis,this results conform with cyprohetadine(antagonist of 5-HT).
