摘要
主要探讨了亚侧耳原生质体制备和再生条件。结果表明,以0.6mol·L-1的NaCl配制成浓度为2%、pH值6.5的溶壁酶溶液,32℃恒温水浴酶解菌龄8d的亚侧耳菌丝体3h,获得原生质体的产量最高。0.6mol·L-1的蔗糖作为再生培养基中的渗透压稳定剂时,再生率最高为6.0×10-2。
The protoplast preparations and regeneration factors of Hohenbuehelia serotina were disscused. The results revealed that the amount of protoplast obtained was the highest in the lysozgme liquor that the concentration was 2% and pH was 6.5, which was made using 0.6 mol·L-1 NaCl. The mycelium of Hohenbuehelia serotina was put into the water-bath at constant temperature 32℃ for 3 h. In addition, the highest regeneration of protoplast of Hohenbuehelia serotina reached 6.0×10-2, when 0.6 mol·L-1 sucrosum was used for stabilizer of osmotic pressure of reproducible medium.
