摘要
目的研究过敏原TNCB对小鼠角质形成细胞(PAM212)促炎症细胞因子IL-18表达的调节效应。方法以不同浓度TNCB处理小鼠角质形成细胞,并以PMA、LPS处理作为阳性对照,利用RT-PCR和ELISA法检测PAM212细胞中IL-18表达变化情况。结果TNCB处理引起角质形成细胞变圆,从支持物脱落;TNCB以剂量依赖的方式促进角质形成细胞内IL-18的mRNA的转录,在处理浓度为10μmol/L时显著提高IL-18 mRNA的转录和蛋白质的分泌。结论过敏原TNCB对角质形成细胞中IL-18表达呈上调效应,为进一步研究炎症反应条件下IL-18的表达调节机制打下了基础。
Objective To investigate the expression changes of pro-inflammatory cytokine IL-18 in murine keratinocytes PAM212 treated with trinitrodichlorobenzene (TNCB). Methods Using phorbol myristate acetate (PMA) and lipopolysaccharide (LPS) as positive control,the IL-18 mRNA expression and IL-18 secretion were detected by RT-PCR and ELISA,respectively. Results Keratinocytes tended to round from polygonal after TNCB treatment; TNCB induced IL-18 mRNA transcription in a dose-dependent manner; TNCB of 10 μmol/L significantly elevated the IL-18 mRNA and protein expressions. Conclusion TNCB can up-regulate IL-18 expression in murine keratinocytes cell line PAM212; this work will benefit the study on regulatory mechanism of IL-18 expression under the condition of inflammatory responses.
