黄芪对肺纤维化大鼠Th1/Th2型细胞因子平衡及自由基代谢的影响 被引量:45

Effects of Astragalus on the balance of Th1/Th2 cytokines and free radical metabolism in rat model of pulmonary fibrosis

摘要 目的研究黄芪水提物、黄芪皂苷对博莱霉素(BLM)所致肺纤维化大鼠Th1/Th2型细胞因子平衡、NO、SOD、MDA及肺组织病理结构的影响,探索黄芪阻抑肺纤维化的效应及其机制。方法Wister大鼠随机分为空白组、BLM模型组、地塞米松组、黄芪水提物组、黄芪皂苷组;气管内注入BLM制作大鼠肺纤维化模型,造模后第2天开始药物干预,14、28d各取6只大鼠,分别检测血IFN-γ、IL-4、NO水平,肺组织SOD、MDA水平及肺组织病理结构的动态变化。结果与空白组比较,模型组大鼠IFN-γ、IFN-γ/IL-4比值、SOD含量降低,IL-4、NO、MDA的含量明显升高,肺组织发生明显的肺泡炎及纤维化改变(P<0.05);与模型组比较,黄芪水提物组、黄芪皂苷组、地塞米松组使大鼠血IFN-γ、IFN-γ/IL-4比值、SOD含量明显升高,NO、MDA含量明显降低,肺组织的肺泡炎及纤维化程度明显降低(P<0.05);黄芪水提物组、黄芪皂苷组与地塞米松组比较,各指标均无统计学差异(P>0.05)。结论黄芪水提物、黄芪皂苷对肺纤维化大鼠肺组织均具有保护作用,调节Th1/Th2型细胞因子的平衡及NO代谢、及提高抗氧化能力可能是其阻抑肺纤维化发生的机理之一。 Objective To observe the effects of Astragalus decoction and Astragalus saponin on the balance of T helper1/T helper2 (Th1/Th2) cytokines,nitric oxide (NO),superoxide dismutase (SOD),malondialdehyde (MDA),and the pathomorphological features in bleomycin (BLM)-induced pulmonary fibrosis rats,and to explore the underlying mechanism. Methods Total of 60 Wister rats were evenly randomized into control,BLM model,Dexamethasone,Astragalus decoction,and Astragalus saponin groups. Pulmonary fibrosis model was established by injection of BLM via trachea (5 mg/kg). After 24 h,rats in the five groups were given corresponding drugs by intragastric administration once daily. At the 14 d and 28 d,6 rats from each group were sacrificed for detecting the levels of Interleukin-4 (IL-4),interferon gamma (IFN-γ),and NO in the serum,as well as the changes of SOD,MDA,and pathomorphological feature in the lung tissue. Results In comparison with blank control group,the levels of IFN-γ and SOD,as well as the ratio of IFN-γ/IL-4 were significantly lower,while IL-4,NO,MDA considerably and the degree of fibrosis were higher in BLM model group (P<0.05); whereas compared with model group,the IFN-γ level,the SOD level,and the ratio of IFN-γ/IL-4 were significantly higher (P<0.05),the levels IL-4,NO,and MDA were significantly lower,as well as the fibrosis degree was more mitigative in Astragalus decoction and Astragalus saponin groups (P<0.05). No significant differences of these indexes were found between Astragalus decoction group,Astragalus saponin group,and Dexamethasone group (P>0.05). Conclusion Astragalus decoction and Astragalus saponin have the inhibitory effects on the process of pulmonary fibrosis,whose mechanisms maybe relate to balancing cytokine,regulating the metabolism of nitric oxide oxidative free radical.
