农杆菌介导仙客来遗传转化体系的建立 被引量:3

摘要 采用根癌农杆菌介导法,研究Mn-SOD基因转化中影响仙客来品种'鲜红1011'叶片转化效率的多种因素,建立仙客来遗传转化体系.结果表明:以叶片为受体材料,培养基中加入50 mg/L Kan(进行抗性筛选),500 mg/L Cef(脱菌),叶片经2 d的预培养,以OD600值为0.4~0.6的菌液侵染5~8 min,共培养3 d有利于仙客来叶片的遗传转化. Several factors that affected genetic transformation of 'scarlet 1011' cyclamen cultivar were examined and mediated by Agrobacteriumtum efaciens. The result showed that the suiltable conditions of transformation contained 50 mg/L Kan,500 mg/L Cef,2 d preculture,0.4—0.6 OD600,5—8 min infection,3 d coculture.Thus a simple and efficient genetic transformation system was developed.
