摘要
以人参愈伤组织细胞悬浮系为材料,研究了人参原生质体的分离纯化方法及影响因素。结果表明:以人参愈伤组织细胞悬浮系为材料,在加入1%纤维素酶和0.5%果胶酶、甘露醇浓度0.7 mol/L的酶解液中酶解8 h分离得到的人参原生质体产量最高;采用人参与胡萝卜共培养培养基,进行看护培养成功地培养出愈伤组织。
Using callus cell suspension of Panax ginseng as materials,protoplast separation and purification of Panax ginseng and their influencing factors were studied.The results showed: when Panax ginseng callus cell suspension was added to the enzyme digestion solution of 1% cellulose,0.5% pectinase and 0.7 mol/L mannitol for 8 h,protoplast production separated from Panax ginseng reached the maximum;Using the carrot of co-culture matrix for nursing training,a callus was successfully developed.
