摘要
目的:探讨大鼠心肺复苏(CPR)后脑功能及病理组织学变化。方法:42只雄性SD大鼠随机分为3组:正常对照组(A组,6只),假手术组(B组,6只),复苏组(C组,按ROSC后1 h、6 h、12 h、24 h、72 h各时点分5亚组,6只)。分别行神经功能缺损评分(NDS),测定脑组织含水量及病理形态学观察。结果:C组大鼠NDS评分降低,随ROSC时间延长,NDS水平逐渐上升,但72 h仍低于正常;大鼠ROSC后脑组织含水量呈上升趋势,24 h达高峰,各时点均高于B组(P<0.01);脑组织存在缺血性病理改变,神经元细胞坏死、胶质细胞增生、血管周水肿、尼氏体减少或消失,以24 h最明显,72 h损伤仍较重;体视学分析显示C组神经元细胞比表面、数面密度下降,平均体积增加(P<0.01)。结论:大鼠CPR后出现神经元细胞坏死、脑水肿、整体行为学评分下降,存在脑功能损害。
Objective:To investigate the change of brain function and pathologic histology in rats after CPR. Method:42 Sprague-Dawley male rats were randomly divided into four groups:normal group (n=6,A),shameoperated group(n=6,B),regular CPR group including 1 h,6 hs,12 hs,24 hs and 72 hs after ROSC(5 sub-groups, n=6 for each,C).At each time point assessed the Neurodeficit Scores(NDS),calculated the brain water content,and pathological change was observed.Results:NDS decreased obviously in CPR group than B group (P < 0.01),and increased obviously along with the prolongation of time following ROSC,but it was still lower than normal.The brain water content in rats of C group increased obviously along with the prolongation of time following ROSC,and reached its peak at ROSC 24 h,much higher than that of B group (P < 0.01).Brain tissue appeared change of ischemia pathology.The neurocell necrosis,gliocyte proliferation,perivascular edema,tigroid body diminished,it was most sever at ROSC 24 h,and still obviously at ROSC 72 h time point.Stereology analytic results found that S-V,NA of nerve cell decreased,VQ increased,manifestly at 24 h (P < 0.01) after ROSC in C group.Conclusions:There is brain injured in rats after CPR,such as neuron necrosis,brain edema and NDS decreased.
