牛尾山药离体再生体系的建立初探 被引量:4

摘要 采用安顺刘官牛尾山药茎段和顶芽为外植体,研究了不同激素对山药再生体系建立的影响.结果表明:以MS为基本培养基幼嫩茎段在MS+6-BA 1.0 mg/L+NAA 0.3 mg/L的培养基中,可直接形成根系发达、健壮的山药组培苗,顶芽在6-BA 4.5 mg/L、NAA 2.0 mg/L的MS培养基中愈伤诱导效果最好,以MS+NAA 0.5 mg/L+6-BA 1.0 mg/L培养基适于丛生芽分化,以0.3 mg/L NAA与0.3 mg/L 6-BA的培养基有利试管苗增殖,生根以加NAA 0.2~0.3 mg/L的MS培养基生根效果最佳,生根率达90%以上. Stem segments and apical buds of Dioscrea batatas in Liuguan Township,Anshun district were used as explants to study the effect of different hormone on the establishment of in vitro regeneration system.The results showed that the young stem segment could develop to healthy plantlet with well-developed root system on MS+6-BA 1.0 mg/L + NAA 0.3 mg/L media based on MS basic media,apical bud had best callus induction in MS + 6-BA 4.5 mg/L + NAA 2.0 mg/L,MS + 6-BA 1.0 mg/L+ NAA 0.5 mg/L was suitable for differentiation for clustered buds,6-BA 0.3 mg/L+ NAA 0.3 mg/L was suitable for plantlet propagation,and MS + NAA 0.2~0.3 mg/L was best for rooting with 90% of rooting rate.
