摘要
目的研究三七三醇皂苷(PTS)、胞二磷胆碱(CDP-c)联用对大鼠局灶脑缺血/再灌注的神经保护作用。方法用大脑中动脉栓塞法制作短暂性(90min)局部脑缺血/再灌注模型,观察PTS、CDP-c单用及联用7d后,大鼠脑梗死体积、caspase-3表达及凋亡细胞数。结果和对照组相比,PTS组、CDP-c组及两药联用组脑梗死体积较小,caspase-3表达、凋亡细胞数均较少,并有统计学意义。两药联用组和PTS组、CDP-c组相比,脑梗死体积较小、caspase-3表达、凋亡细胞数较少(P<0.05)。PTS组和CDP-c组相比各指标无统计学意义。结论PTS和CDP-c单用对短暂脑缺血/再灌注模型可能具有神经保护作用。PTS与CDP-c联用对短暂性局造脑缺血/再灌注的神经保护可能有协同作用。
Objective To investigate the neuroprotective effects of PTS and CDP-c in rat model of transient focal cerebral ischemia-reperfusion. Methods The model of acute reperfusion injury after transient cerebral ischemia in rat was made by middle cerebral artery occlusion. Expression of caspase-3 and apoptotic cells in infarction were detected, and infarct volumes were assessed after PTS, CDP-c and their combined administration for 7 days. Results Compared to placebo group, infarct volume of PTS,CDP-c and combination groups were significantly reduced,and the number of caspase-3 positive cells and apoptotic cells were significantly fewer as well,respectively(P<0.05).meanwhile compared to both PTS group and CDP-c group infarct volume of combination group were significantly reduced,and the number of caspase-3-positive cells and apoptotic cells were significantly fewer, respectively. However, there was no significant difference between PTS group and CDP-c group. (P>0.05). Conclusion PTS and CDP-c may be respectively exert a neuroprotective effect during the transient experimental focal cerebral ischemia-reperfusion model.The results of this article demonstrates a possible synergistic neuroprotection effect of PTS and CDP-c after transient experimental focal cerebral ischemia-reperfusion.
