摘要
目的探讨血管内皮细胞生长因子(VEGF)基因936C/T位点多态性在中国汉族人群中的分布及其与动脉粥样硬化性脑梗死(ACI)的关联。方法应用PCR-限制性片段长度多态性技术检测155例ACI患者和195例健康对照者的936C/T位点多态性,以非条件Logistic回归校正混杂因素,并进行多态性与ACI患者以及其他相关或者风险因素分层的统计学分析。结果936C/T的多态性位点的基因频率分布符合Hardy-Weinberg平衡定律;VEGF基因936位点C/C与C/T+T/T之间基因型频率在两组人群中的比较差异有统计学意义,P<0.001,OR=2.34;分层分析发现:吸烟或患高血压、糖尿病的患者中其易感性更为显著(均P<0.001)。结论VEGF基因936C/T多态性与ACI有着明显的易感关联,而吸烟和高血压、糖尿病加强此种易感性关联。
Objective To investigate the distribution of vascular endothelial growth factor (VEGF) gene 936 C/T polymorphism in Chinese Han population and the relationship between the polymorphism and atherosclerotic cerebral infarction (ACI).Methods Peripheral blood samples were collected from 155 patients with ACI, 91 males and 64 females, aged 67.01±9.83, and 195 gender, age smoking status, and drinking status matched healthy controls, 118 males and 77 females, aged 65.20±9.09. PCR-restriction fragment length polymorphism technique was used to detect the 936 C/T polymorphism. Unconditional logistic regression was used to adjust the intermingling factors. Stratified analysis was conducted to analyze the association of ACI with the relevant factors. Results The distribution of 936C/T gene polymorphism was in accordance with Hardy-Weinberg equilibrium. The C/T and T/T genotype frequencies in the ACI group were significantly higher than those in the control group (P<0.001,OR=2.34). The subgroup analyses revealed that the association of the 936 C/T polymorphic genotypes in the smoking ACI patients and those with hypertension and/or diabetes mellitus were significantly higher than in the non-smoking ACI patients and those without hypertension and/or diabetes mellitus(all P<0.001). Conclusion 936 C/T polymorphism of VEGF gene is a predisposing factor of ACI in Chinese Han population. Smoking custom, hypertension, and diabetes mellitus will enhance the risk of ACI in the individuals with 936C/T polymorphism.
