摘要
以农大钙果3、4、5号品种为研究对象,研究其组织培养及快速繁殖技术。结果表明:①以钙果幼嫩茎段的侧芽为外植体,消毒效果最好。②以MS为基本培养基,初代培养附加0.4mg/L6-BA和0.4mg/LIAA腋芽萌发最好;增殖培养附加0.4mg/L6-BA和0.5mg/LIAA腋芽增殖效果最佳。③生根培养阶段,以1/2MS为基本培养基,附加0.5mg/LIBA生根效果最好。④选用山土作为钙果试管苗的移栽基质效果最佳。
Experiment was carried out using three varieties of Cerasus humilis Bunge as material for studies of its tissue culture and rapid propagation techniques.The results showed that: ①The disinfection effects was the best using the lateral bud of young stems as explant.②The medium MS+6-BA 0.4 mg/L+ IAA 0.4 mg/L+30 g/L sugar+6 g/L agar was the optimal one for germination,and the medium MS+6-BA 0.4 mg/L+IAA 0.5 mg/L+30 g/L sugar+6 g/L agar was the optimal one for differentiation. ③The effective medium for rooting was 1/2MS+IBA 0.5 mg/L+30 g/L sugar+6 g/L agar. ④The best transplanting base material was mountain soil.
