摘要
目的探讨罗格列酮辛伐他汀对兔动脉粥样硬化形成的影响。方法40只日本大耳白兔随机分为五组,正常对照组、高脂模型组、罗格列酮组、辛伐他汀组、罗格列酮+辛伐他汀组,每组8只。A组(正常对照组):普通颗粒饲料喂饲150g,日二次,10周。B组(高脂模型组):高脂饲料(1%胆固醇+8%猪油+普通颗粒饲料)150g,日二次,10周。C组(罗格列酮组):高脂饲料喂饲10周,从喂饲高脂饲料起,给予口服罗格列酮0.5mg/kg/日。D组(辛伐他汀组):高脂饲料喂饲10周,从喂饲高脂饲料起,给予口服辛伐他汀2.5mg/kg/日。E组(罗格列酮+辛伐他汀组):高脂饲料喂饲10周,从喂饲高脂饲料起,给予口服罗格列酮0.5mg/kg/日+辛伐他汀2.5mg/kg/日。实验结束后获取血浆和主动脉全长标本进行生化和病理分析。结果罗格列酮组、辛伐他汀组、罗格列酮+辛伐他汀组三组C-反应蛋白(CRP)明显低于高脂模型组P<0.01,罗格列酮组、罗格列酮+辛伐他汀组非对称二甲基精氨酸(ADMA)血浆水平低于高脂模型组P<0.01,罗格列酮组、辛伐他汀组、罗格列酮+辛伐他汀组三组斑块面积占纵剖面主动脉面积之比明显小于高脂模型组。结论过氧化物酶增殖物激活受体激动剂罗格列酮以及他汀类药物辛伐他汀通过抑制炎症反应可以防治动脉硬化的发生发展,具有协同作用。
Objective To study the effect of rosiglitazone on atherosclerosis and potential mechalism in rabbits.Methods Fourty healthy Japanese big-ear rabbits were randomly divided into five groups with 8 rabbits in each group:normal control group,model group,rosiglitazone group,simvastatin group,rosiglitazone and simvastatin group.Group A were fed with common forage for 10 weeks;group B were fed with fat forage(common forage contained 1% cholesterol and 8% lard);group C were fed with fat forage and rosglitazone 0.5/kg/day;group D were fed with fat forage and samvastatin 2.5/kg/day;group E were fed with fat forage and rosglitazone 0.5/kg/day and samvastatin 2.5/kg/day.At the end of the experiment,the rabbit blood samples were collected for biochemistryl analysis and aorte were prepared for morphologic analysis.Results The level of CRP in group C,group D,group E were significantly greater than that of model group.The level of ADMA were lower in group A,group C and group E than in group B and group D.Group C,group D and group E reduced the extend of atherosclerosis of longitudinal section.Conclusion Rosiglitazone and Samvastatin can prevent the development of atherosclerosis through inhibiting the inflammatory reaction and the effect of them were synergetic.
