雾化吸入BCG对预防哮喘小鼠气道炎性反应的初步研究 被引量:2

摘要 目的探讨卡介苗(BCG)雾化吸入对变应性哮喘的预防作用及其与支气管肺泡灌洗液(BALF)中γ-干扰素(IFN-γ)的关系。方法昆明小鼠40只,随机分成3组,正常对照组、哮喘模型组及预防组。预防组又分为致敏前BCG预防组,激发时BCG预防组和地塞米松预防组。预防组经处理后,与哮喘模型组期制作哮喘模型。观察小鼠气道壁炎症变化,并收集支气管肺泡灌洗液(BALF),测定BALF中细胞总数,嗜酸性粒细胞和淋巴细胞数,用酶联免疫法测定BALF中IFN-γ的含量。结果哮喘模型组BALF中细胞总数和嗜酸细胞数显著高于正常对照组(P<0.01),BALF中IFN-γ水平显著低于正常组(P<0.01);致敏前BCG预防组、地塞米松预防组BALF中细胞数、EOS细胞数显著低于哮喘组,而IFN-γ水平显著高于哮喘组(P<0.01-0.05)。激发时BCG预防组各项指标与哮喘组相比无明显变化。结论雾化吸入BCG能明显抑制哮喘小鼠气道的炎性反应,早期吸入BCG对哮喘小鼠有预防作用。 Objective To study the preventive effect of bovis-Bacillus Calmette-Guérin (BCG) fog-inhalation on allergic asthma in mice and the relation with IFN-γ levels in BALF.Methods 40 KUNMING mices were randomly divided into three groups:normal group,asthmatic model group,preventive group.The preventive group was randomly divided three groups again:presensitization BCG preventive group,challenging BCG preventive group and dexamethasone preventive group.The preventive group after deal,asthmatic model group were made in allergic asthma model in OVA.Pathological scores,bronchoalveolar lavage(BAL) total cell count and differentiation were studied.IFN-γlevels in BAL fluid (BALF)were detected by ELISA.Results the number of BALF total cell count and eosinophils in asthmatic model group were more than in the normal group (P<0.01),IFN-γ level in BALF was decreased(P<0.01);In presensitization BCG prevention group and dexamethasone prevention group the number of BALF total cell count and eosinophils were significantly lower than asthma group,and the IFN-γ level significantly higer than asthma group(P<0.01-0.05).These parameters in challenging BCG prevention group were not significantly changing compared with asthmatic model group(P>0.05).Results BCG fog-inhalation can not only markedly inhibit the airway inflammation in allergic asthmatic mice,but also BCG vaccination in early life prevent in allergic asthmatic mice.
