摘要
目的通过检测鹅脱氧胆酸(CDCA)刺激下,小鼠巨噬细胞中细胞间黏附分子-1(ICAM-1)的表达变化,探讨胆汁酸对巨噬细胞中相关炎性反应分子表达的影响。方法采用反转录-聚合酶链式反应(RT-PCR)及Western blot测定在不同浓度CDCA刺激下,小鼠巨噬细胞株RAW264.7中ICAM-1的mRNA和蛋白水平的变化情况。结果同对照组相比,在CDCA浓度为25、50μmol/L的刺激下,ICAM-1的mRNA和蛋白表达明显升高(P<0.05),两者的表达变化趋势较一致。结论在CDCA的刺激下,小鼠巨噬细胞中ICAM-1的表达明显增加,对于CDCA作为药物在临床上使用的作用研究方面具有重要意义。
Objective To investigate the effect of chenodexycholic acid(CDCA)on the expression of intracellular adhesion molecule-1(ICAM-1) in RAW264.7 cells and evaluate the effect of bile acid on inflammation.Methods RAW264.7 cells were cultured and treated with CDCA. The mRNA level of ICAM-1 was detected by reverse transcription-polymerase chain reaction (RT-PCR), western blot was used to assess the expression of ICAM-1 protein level. Results CDCA significantly increase ICAM-1 mRNA and protein level in RAW264.7 cells (P<0.05) in a concentration-dependent manner. Conclusion CDCA treatment increase ICAM-1 expression in murine macrophage cell line.
