沙丁胺醇喷雾剂对致敏豚鼠和大鼠气管平滑肌痉挛的显效关系研究

The dose-effect relationgship studies of Salbutamol on allergized guinea pigs and the spasm of tracheal smooth muscle in rats

目的:研究沙丁胺醇喷雾剂对过敏性支气管平滑肌痉挛的缓解作用。方法:采用离体气管法和肺溢流法观察沙丁胺醇喷雾剂对致敏豚鼠和大鼠的支气管平滑肌痉挛的缓解作用,采用整体动物药物引喘法观察沙丁胺醇喷雾剂对致敏豚鼠过敏性哮喘保护作用。结果:沙丁胺醇10-6molL、3×10-6molL、10-5molL剂量下,对致敏豚鼠离体气管平滑肌收缩有抑制作用;0.2mgkg、0.4mgkg、0.8mgkg沙丁胺醇对致敏麻醉大鼠肺溢流量具有抑制作用;该剂量下沙丁胺醇对致敏豚鼠过敏性哮喘有保护作用,均产生显著性差异。结论:沙丁胺醇喷雾剂对致敏豚鼠离体气管平滑肌有松弛作用,明显延长组织胺与乙酰胆碱致敏豚鼠引喘潜伏期,减少致敏麻醉大鼠的肺溢流量。

Objective:To study the relief effects of Salbutamol(SAB)spray on anaphylactic bronchus smooth muscal spasm.Methods:To observe the relief effects of SAB spray on bronchus smooth muscle spasm in allergized guinea pigs and rats by isolated guinea pigs sheets and Konzett and Rossler experiments and the protective effects of SAB spray on allergic asthma in allergized guinea pigs by the whole animal experiment.Results:At the doses of 10-6mol L?3×10-6mol L?10-5mol L,There were inhibited effects on the anastole of the isolated tracheal smooth muscle in allergized guinea pigs;at the doses of 0.2mg kg?0.4mg kg?0.8mg kg,SAB could inhibited the lung outflow volume in anesthetized rats which were allergized,and at the above doses,SAB has the protective effects on allergic asthma in allergized guinea pigs.There were significant difference between all above results.Conclusion:Salbutamol spray have the relax effects on tracheal smooth muscle in allergized guinea pigs ex vivo.SAB could lengthen the latent period of asthma induced by histamine and acetylcholine in guinea pigs,and reduce the lung outflow volume in anesthetized rats which were allergized.
