The Effects of the Extractive Flavone of Ovateleaf Actinidia Leaf on Histopathology and Ultrastructural Pathology in Rats after Focal Cerebral Ischemia

摘要 目的研究提取物黄酮对局灶性脑缺血大鼠损伤组织病理及其超微结构的影响。方法Nagasawa线栓法将大鼠制成局灶性大脑中动脉缺血模型。将Wistar雄性大鼠75只随机分为:假手术组、盐水对照组、小剂量给药组、中剂量给药组、大剂量给药组,每组15只。小剂量给药组、中剂量给药组、大剂量给药组于术前30min及术后2h分别给予不同剂量狗枣猕猴桃叶提取物黄酮腹腔内注射。应用TTC染色及图像分析系统测定局部脑缺血24h后的梗死灶体积,HE染色及透射电镜观察大鼠局灶脑缺血后组织病理及超微结构的变化。结果与盐水对照组相比,药物干预组局部缺血后在相同时间内梗死灶的体积较小(P<0.05),超微结构改变也较轻,在实验剂量范围内其病理变化与给药剂量呈负相关。结论狗枣猕猴桃叶提取物黄酮对缺血后大鼠神经元形态和超微结构具有保护作用。 Objective To investigate the effects of the extractive f lavone of Ovateleaf actinidia leaf on histopathology and ultrastructural pathology in rats after focal cerebral ischemia. Methods We induced focal cerebral ischemia model in rats by Nagasawa occlusion methods of middle cerebral artery. Seventy-five healthy Wistar male rats were chosen and randomly divided into sham operated (group A), control group of sodium chloride (group B), little dosages (group C), middle dosages (group D), large dosages (group E). The rats of group C,D,E were given different dosages of the extractive flavone of Ovateleaf actinidia leaf by intraperitoneal injection 30 min before the operation and 2 hours after the operation. We measured the volume of the infarction focus by triphenyl tetrazolium chloride(TTC) stain and image analysis system, and observed the histopathology and ultrastructural pathology by hematoxylin eosin(HE) stain and transmission electron microscope. Results The infarction focus volumes of the groups given the drug were smaller (P<0.05), and the changes of the ultramicrostructure were also lighter compared with the control group of sodium chloride. The pathological change in experiment dose extent had negative correlation with the dose. Conclusion It indicates that the extractive flavone of Ovateleaf actinidia leaf can protect the neurons and the ultramicrostructure after the ischemia.
