摘要
研究了灵芝子实体水提液不同沉淀分离多糖的工艺。结果表明:醇沉法收率高,但需要将灵芝子实体水提液浓缩至小体积,以提高收率、减少乙醇用量;氢氧化钙沉淀法不对水提液浓缩,但要达到较高的收率需要大量的沉淀剂氢氧化钙;采用经过改进的氢氧化钙沉淀法,不对水提液浓缩,通过加入沉淀剂聚合硫酸铁,大幅度减少了氢氧化钙的加入量,多糖沉淀分离完全,收率与醇沉法相近。
Isolation techniques of polysaccharides of Ganoderma Lucidum from water-extracted solution(polysaccharides solution extracted from the fruiting body of ganoderma lucidum in water)are investigated including ethanol precipitation,calcium hydroxide precipitation and improved calcium hydroxide methods. The results show that ethanol precipitation method have high yield of polysaccharides. In order to increase the yield and decrease consumption of ethanol,water-extracted polysaccharides solution must be concentrated to a small volume. Calcium hydroxide precipitation method can be used to have high polysaccharides yield without enrichment of water-extracted polysaccharides solution,but consumption of calcium hydroxide is very high. In order to get high polysaccharides yield and simplify isolation process,the improved calcium hydroxide precipitation method was developed with the addition of PFS,This method has advantage of decreasing the consumption of calcium hydroxide,and the polysaccharides yield is similar to that of ethanol precipitation method.
