几种黏膜免疫佐剂对小肠树突状细胞、MHCⅡ类分子及核转录因子NF-kB的影响 被引量:2

摘要 目的研究黏膜免疫佐剂在小肠局部的抗原递呈方式和信号转导途径。方法应用大鼠肠道原位结扎灌注模型观察大豆黄酮、CpGDNA和乳酸杆菌3种黏膜免疫佐剂对树突状细胞、MHCⅡ类分子和核转录因子NF-κB的影响。结果1)CpGDNA和乳酸杆菌能够显著增加小肠树突状细胞的面积,促进局部疫苗抗原的递呈;2)大豆黄酮、CpGDNA和乳酸杆菌能在24h内促进大鼠空肠MHCⅡ类分子的显著表达。提示黏膜免疫佐剂可能通过增强黏膜局部抗原递呈细胞的活性和MHCⅡ类分子的含量促进局部特异性免疫反应;3)CpGDNA可以在24h内显著增加细胞内NF-κB蛋白的含量,在0.25h作用最明显;乳酸杆菌在6h内可极显著增加NF-κB蛋白的表达;而大豆黄酮对细胞内NF-κB蛋白的含量作用不明显。结论提示细菌类佐剂CpGDNA和乳酸杆菌是通过活化NF-κB途径激发免疫活性细胞,大豆黄酮可能不通过该途径。 Objective To study the way of presenting antigen and pathway of adjuvants in activating immune system of intestine.Methods The effects of daidzein,CpG,and lactic acid bacteria(LAB)on dendritic cells(DC),MHC II molecule,and nuclear factor-κB(NF-κB)were determined by using model rats ligated and perfused intestinal segments.Results 1)CpG and LAB could significantly increase the numbers of DC in intestine,which suggested that adjuvants could activate DC,promote antigen presenting,and induce specific immune res...
