氯胺酮对小鼠离体十二指肠纵行肌条收缩的影响

摘要 目的:探讨氯胺酮对小鼠离体十二指肠纵行肌条收缩的影响及作用机制。方法:取小鼠十二指肠纵行肌条,放入盛满Kreb’s液的灌流肌槽中,分别加入不同浓度的氯胺酮,记录其加药前后十二指肠纵行肌条收缩活动变化;观察10-4mol/L氯胺酮与相关工具药(乙酰胆碱、阿托品、氨茶碱、L-NNA)对十二指肠纵行肌条收缩活动的影响。结果:氯胺酮可明显抑制十二指肠肌条的收缩;阿托品及氨茶碱均可增强氯胺酮对十指肠肌条收缩的抑制效应(t=1.92,1.94,P<0.05);且氯胺酮可拮抗乙酰胆碱对十二指肠肌条收缩的促进作用(t=1.88,P<0.05);加入L-NNA后,其对氯胺酮抑制十二指肠肌条收缩的作用无影响(t=1.02,P>0.05)。结论:氯胺酮对小鼠离体十二指肠纵行肌条的收缩有明显抑制作用;机制可能是通过阻断乙酰胆碱与小肠壁上M2受体的结合,抑制Gi/Go亚单位,提高细胞内的cAMP含量,从而抑制消化道平滑肌运动。 Aim:To analyze the effect and mechanism of ketamine on contraction of isolated duodenal longitudinal strips in mice.Methods:The strips derived from mice were placed in a Kreb’s perfusion muscular socket,and ketamine and other relevant reagents were added so as to record the changes of its contraction activity.Results: Ketamine could apparently inhibit the contraction of the duodenal strips;atropin and aminophylline both could enhance ketamine’s inhibitory effect(t=1.92,1.94,P<0.05).Ketamine could also antag...
