卵巢上皮性癌组织中PTEN蛋白与mRNA的表达

Expression of PTEN protein and mRNA in ovarian epithelial carcinoma tissue

目的:检测卵巢上皮性癌组织中PTEN蛋白和mRNA的表达。方法:应用免疫组织化学SP法检测12例正常卵巢组织,20例卵巢良性上皮性肿瘤组织,65例卵巢上皮性癌组织中PTEN蛋白的表达。采用RT-PCR法检测25例卵巢上皮性癌及12例正常卵巢组织中PTENmRNA的表达。结果:PTEN蛋白在卵巢上皮性癌组织中的阳性表达率为44.6%,正常卵巢组织为100%,卵巢良性肿瘤组织为85.0%,3者之间差异有统计学意义(χ2=19.584,P<0.05)。卵巢上皮性癌组织与卵巢良性和正常卵巢组织之间比较差异有统计学意义(χ2=12.482、10.046,P<0.05)。PTEN蛋白的表达与临床分期和组织学分级(χ2=6.232、5.071,P<0.05)。正常卵巢组织中PTENmRNA的相对表达量高于卵巢上皮性癌(1.30±0.32vs0.61±0.20,t=6.706,P<0.05)。结论:PTEN表达缺失参与卵巢上皮性癌的发生发展。

Aim:To analyse the expression of PTEN protein and mRNA in ovarian epithelial carcinoma tissue.Methods:The expressions of PTEN in 12 cases of normal ovarian tissues,20 cases of benign tumors and 65 cases of ovarian epithelial cancer tissue were detected by immunohistochemical method.
