摘要
目的:探讨蓝氏贾第鞭毛虫(简称贾第虫)在分裂过程中是否形成典型的凝集染色体以及染色体的数目。方法:用改良的TYI-S-33培养基培养贾第虫滋养体。分别在不同浓度的秋水仙素(0、0.05、0.10、0.20、0.40和0.80mg/L)、低渗处理不同时间(30、50、55、65和80min)和不同固定液[V(甲醇):V(冰醋酸)为3:1和2:1]等条件下制备贾第虫染色体。结果和结论:在制备贾第虫染色体时,秋水仙素的最佳作用浓度为0.20mg/L,最佳低渗处理时间为55min,固定液推荐V(甲醇):V(冰醋酸)为3:1。贾第虫在分裂过程中形成典型的凝集染色体,染色体数目为2n=10。
Aim:To study whether the Giardia lamblia have typical chromosomes and the chromosome number during mitosis.Methods:Trophozoites of Giardia lamblia were grown in modified TYI-S-33 medium at 37 ℃.The medium was removed and cell cultures were enriched with fresh,warmed medium.The chromosome preparation were reseached from content of colchicines(0,0.05,0.10,0.20,0.40,and 0.80 mg/L),time of low osmosis(30,50,55,65,and 80 min)and the kind of fixation fluid[V(carbinol):V(glacial acetic acid)was 3:1 or 2:1].Results...
