摘要
目的 观察研究大鼠椎间盘软骨终板的细胞形态及表型特征 ,建立椎间盘细胞培养的体外模型。方法 酶消化法培养大鼠椎间盘的软骨终板细胞 ,HE和甲苯胺蓝染色 ,进行光镜及电镜观察 ;使用免疫组化法 (SP)检测椎间盘软骨细胞的特征性II型胶原表达情况 ,与同种动物不同部位的软骨细胞做对照。结果 HE染色见细胞呈多角形 ,核为圆形或椭圆形 ,有时见有双核 ,胞浆内可见空泡。甲苯胺蓝染色可见胞浆及细胞周围有紫红色或红色异染出现。电镜观察见细胞胞浆内有丰富的粗面内质网系统及线粒体 ,核呈圆形或椭圆形 ,核膜明显 ,细胞膜下及胞浆中有较多的无界膜空泡。原代细胞与传代细胞之间形态和结构无明显差别。软骨终板和关节软骨细胞形态相似。大鼠椎间盘软骨终板细胞表达II型胶原。结论 本研究提示软骨终板细胞表达软骨细胞的特征性胶原 。
Objective To observe the morphology and phenotypes of cells extracted from the intervertebral discs of young rats, establish the intervertebral disc chondrocyte lines. Methods The intervertebral disc endplate of rats were digested enzymatically. The morphology of those cells was examined by light microscopy and transmission electron microscopys. The special collagen II was determined by immunhistochemical analysis. At the same time, to compare with the ariticular chondrocytes from the same animals.Results The shape of the primary cells appear as around and oval.The cytoplasm was blue after stained with the toluidine blue.The morphology of the cartilage endplate cells and the articular cells was almost identical under the transmission electronic microscope. All freshly extracted cells expressed collagen II. Conclusion Findings showed that intervertebral disc endplate cells express cartilage-specific matrix proteins, like the articular cartilage cells do. This experiment forms a foundation of cell biology for the degradation of the disc.
