摘要
目的探讨白细胞介素-6(IL-6)对大鼠脊髓损伤后神经细胞凋亡的影响。方法SD大鼠随机分为3组,IL-6治疗组(A组),单纯脊髓损伤(SCI)(B组),假损伤组(C组)。每组20只动物,应用改良Allen打击法制作急性SCL模型,A组SCL后30min,腹腔内注射10μgIL-6,B组单纯脊髓损伤,无治疗,C组,暴露硬脊膜,无SCL,术后分别12h和12周,每组10只动物,取脊髓伤段标本测小离子含量和Bcl-2、Fas蛋白表达的测定(免疫组化法)。结果(1)SCL后组织水肿,Na+、Ca2+离子浓度升高,K+,Mg2+离子浓度降低。IL-6治疗后显著改善这些变化。(2)A、B、C中,均发现Bcl-2和Fas蛋白阳性表达,表达顺序分别为A>B>C和B>C>A。结论SCI后早期应用IL-6可抑制大鼠脊髓细胞凋亡,减少神经细胞离子失衡,改善细胞内环境,从而对脊髓损伤起到保护作用。
Objective To study the effccts of the interlcukin -6(IL -6)on rat neurons apoptosis after spinal cord injury (SCI ). Methods The 60SD mice were divided into three groups :IL -6group (group A ),SCI (Group B )and sham group (group C, on SCI ).12h after operation ,10 rats of each group were kiled and injured segment of spinal cord wsa resecled to be examined with atomic absorption speclropjpt, eru .At 12weeks after operation the another 10 rats were exanined Bel -2and Fas protein expression (inmunohistochemically ). Results There was a signifieant incerase of water contant and Na+、Ca2+ions and decrease of K+,Mg2+ions in the injured eord segment of group B .the inuection on IL -6group improvedthe neurologieal ouleome of injured spinal cord. The strong experssion sequcnce of Bcl-2and Fas protein experssion were found in all three groups, but A>B>and B>C>A(P < 0.05)repectively. Conclusions The early adminstration of IL -6 could be inhibit apoptosis and improvement of the intenalion enviroment of after spinal cord injury.
