摘要
目的 研究碱性成纤维细胞生长因子( Basic Fibroblast Growth Factor, bFGF)对成骨细胞与多孔生物材料粘附特性的影响。 方法 分别用 0.1、 1、 10、 100、 1 000 ng/ml系列浓度的 bFGF诱导培养基诱导培养成骨细胞 24 h后,接种成骨细胞于多孔生物材料上,测定接种后 1 h的粘附细胞率。结果 bFGF在 0~ 10 ng/ml范围内能促进细胞在材料上的粘附,并在 10 ng/ml浓度下达到最大粘附率( P< 0.05),当 bFGF浓度进一步增加时,细胞粘附率不再增加,在 1 000 ng/ml时细胞粘附率与对照组无明显差异。结论 10 ng/ml的 bFGF诱导可明显促进成骨细胞与多孔生物材料间的粘附。
Objective To investigat the influence of basic fibroblast growth factor on the adhension characteristics of osteoblasts.Methods 0.1,1,10,100,1 000 ng/ml bFGF were used to induce osteoblasts came from rabbit bone marrow stromal cells for 24 hours. The attached cells was calculated with fluorescence method at 1 h after seeding . Result The number of attached cells had increased after the cells were induced by bFGF when the concentration below 10 ng/ml, and reached the highest number when induced by 10 ng/ml bFGF (P< 0.01). While the number didn't increase with the increase of bFGF concentration. There were no differences when induced by 1 000 ng/ml bFGF compared with the control group. Conclusion The induction of 10 ng/ml bFGF can significantly promote the adhension of osteoblasts to porous biomaterial.
