摘要 目的探讨成纤维细胞生长因子( bFGF)和重组人骨形成蛋白- 2( rhBMP- 2)单独和联合作用对骨髓基质细胞( BMSC)的增殖和分化的影响。方法利用四唑盐比色法( MTT),细胞总蛋白考马斯亮蓝测定法,碱性磷酸酶( ALP)试剂盒分别测定不同浓度的 bFGF和 rhBMP单独和联合作用后 BMSC的增殖和分化情况。结果 bFGF单独作用促进 BMSC的增殖,但高浓度抑制 ALP表达, rhBMP对 BMSC的增殖和 ALP和总蛋白含量均有促进作用,而 bFGF和 rhBMP联合作用后 BMSC的增殖和 ALP活性较单独作用有更明显的升高。结论 bFGF和 rhBMP联合作用对于促进 BMSC的增殖和向成骨细胞分化有协同作用。 Objective To investigate the effects of basic fibroblast growth factor (bFGF) and recombinant human bone morphogenetic protein- 2 (rhBMP- 2) on the proliferation and differentiation of bone marrow stromal cells (BMSCs).Method BMSCs were subjected to the treatment with different concentrations of bFGF, rhBMP- 2, bFGF and rhBMP- 2, after which the attached cells were measured with methylthiazol tetrazolium (MTT) method, total cellular protein method and alkaline phosphatase (ALP) measurement kit.Result bFGF alone could promote the proliferation of BMSCs. Significantly greater proliferation and differentiation were found in combined group comparing with individual groups. Conclusion bFGF and rhBMP- 2 might promote the proliferation and differentiation to osteoblast of BMSCs and have synergistic action.
