创伤修复中细胞信号传递对瘢痕形成的影响:SH2功能领域蛋白酪氨酸磷酸酶的表达及意义 被引量:4

摘要 目的观察瘢痕与正常组织上皮细胞中SH2功能域蛋白酪氨酸磷酸酶(SH-PTH2)的表达特征,探讨创伤修复过程信号途径信号与瘢痕形成的影响。方法选取特异性SH-PTH2抗体,利用免疫组织化学技术对临床收集到的创作后不同时期的6例瘢痕组织标本及同体6例正常皮肤标本进行染色,光镜下观察。结果瘢痕组织与正常皮肤的皮肤角质形成细胞中SH-PTH2抗体的表达存在明显差异,增生性瘢痕皮肤中SH-PTH2高于正常皮肤对照组。同时在瘢痕形成的不同时期内,真、表皮SH-PTH2免疫阳性的表达差别。结论在皮肤伤口的愈合过程中,SH-PTH2对创面修复过程中信号转导的调节作用,可能影响着愈合的结局。 Objective To investigate the expression of SH-PTP2 in cutaneous cells of hyperplastic scar formation in the process of wound repair.Method Immuno-histochemistry staining with specific antibody was employed to determine the expression of src homology domain-protein tyrosine phosphatases(SH-PTP2) in skin of hyperplastic scars(n=6) and normal human(n=6).Result There was an evident difference in SH-PTP2 positive keratinocytes between hyperplastic scars and normal skin.The SH-PTP2 antibody expression in hyperplastic scars was much stronger than that in normal skin.There were a lot of immuno-reaction positive cells in each stage of scar formation,and the intensity of expression was stronger in proliferative stage than that in mature stage in dermis.There was no significant difference in SH-PTP2 expression in epidermis between proliferative stage and mature stage.Conclusion Keratinocyte proliferation and migration are essential to cutaneous wound healing and are mediated by growth factor receptor/ligand interactions.Stimulation of growth factor receptors by ligand(s) may lead to activation of signaling molecules including SH-PTP2,Hence,SH-PTP2 may influence signal transduction regulation in wound repair.
