皮肤溃疡伤口中Bax和Bcl-2蛋白含量的变化及其与溃疡发生的关系 被引量:24

摘要 目的观察Bax和Bcl-2蛋白在溃疡、溃疡边缘和正常皮肤组织中的表达特征及其对溃疡形成的影响。方法24例被测标本中包括不同类型的溃疡组织8例,其对应的溃疡边缘和正常皮肤组织各8例。用常规病理技术和免疫组化方法确定这两种蛋白在溃疡组织和正常皮肤中的定位和表达量的变化规律。结果在正常皮肤组织中,Bax蛋白的阳性颗粒较少,在表皮细胞内稍有表达,而Bcl-2蛋白则主要定位于表皮细胞和部分成纤维细胞中。在溃疡组织中,Bax蛋白存在于表皮基底层细胞和部分成纤维细胞的胞浆中,而Bcl-2蛋白在表皮基底层细胞内有表达。从正常皮肤到溃疡组织,Bcl-2含量逐渐下降,而Bax的蛋白表达呈升高趋势。结论在正常皮肤和溃疡组织中,Bax和Bcl-2蛋白不同分布和表达变化规律显示,Bcl-2和Bax两种蛋白可能与溃疡创面处细胞凋亡增加相关,从而导致伤口愈合迟缓和溃疡的发生。 Objective To investigate the difference in protein contents of Bax and Bcl-2 in ulcerative and normal skins and to explore their effects on ulter formation.Method Twenty-four cases were detected to compare the expression and distribution of Bax and Bcl-2 with normal pathological methods and immunohistochemistry method. Among those cases, there were 8 cases of ulcerative tissues, 8 cases of ulcer edge skins, 8 cases of normal skins.Result Positive signals of Bax and Bcl-2 could be found in normal skins. Bax was found to be located in the epidermal cells. Bcl-2 could be detected in epidermal cells and some fibroblasts. In ulcerative tissues, Bax positive granules could be detected in epidermal cells and some fibroblasts, while Bcl-2 protein was only existed in cytoplasm of epidermal basal cells. From normal skins, ulter edge to ulcers, the content of Bcl-2 was decreased progressively.while the protein expression of Bax was increased gradually .Conclusion The different expression and distribution of Bax and Bcl-2 in uclerative versus normal skins might be associated with the mechanism controlling cell apoptosis which lead to ucler formation.
