补阳还五汤药物血清对周围神经再生的影响

Effects of the rat's serum with Bu Yang Huan Wu Tang on peripheral nerve regeneration

目的用血清药理学方法观察补阳还五汤对体外培养的施万细胞生长情况的影响。方法用补阳还五汤药物血清及对照组血清培养取自Wistar新生鼠的施万细胞,在种植不同时间时观察各组的生长情况:用S-100免疫组织化学染色的方法来鉴定所取施万细胞的纯度,同时,用MTT法测定各组的OD值,推测细胞的活性。结果(1)免疫组织化学染色结果显示:所取细胞中85%以上是施万细胞,达到了试验所需要求;(2)药物组细胞比对照组细胞生长形势好,突起长度明显长于对照组(P<0.05);(3)药物组OD值明显高于对照组(P<0.05),而且有一定的血清浓度依赖性。结论本实验成功地建立了中药促进周围神经再生的血清药理学实验方法,并用此实验方法证明了补阳还五汤对周围神经再生的促进作用,为进一步阐明补阳还五汤治疗周围神经损伤的机制打下了基础。

Objective To investigate the effects of the rat's serum with Bu Yang Huan Wu Tang on Schwann cells in culture.Method (1)Cultured Schwann cells with the serum with Bu Yang Huan Wu Tang.(2)Measured the length of the processes of the processes of all groups at different time.(3)S-100 immunhistochemical staining were performed to find the percentage of Schwann cells.(4)Applied MTT assay to reveal OD value of all groups.Result (1)The processes of experimental groups were significantly larger those of control groups(P< 0.05).(2)S-100 immunhistochemical staining showed that more than 85% cells in culture are Schwann cells.(3)MTT colorimetric assay revealed experimental groups elevated OD value compared with control groups(P< 0.05).Conclusion Using the serum pharmacology method,we verified that Bu Yang Huan Wu Tang can promote peripheral nerves regeneration.This research laid a foundation for further studying the mechanisms of Chinese herbs healing peripheral nerve lesion.
