血液透析患者Fas/FasL介导的外周血淋巴细胞凋亡 被引量:1

摘要 目的 探讨Fas/FasL介导的外周血淋巴细胞凋亡对血液透析(血透)患者免疫机能损伤影响。方法 酶联免疫夹心法检测35例血透患者血浆可溶性Fas、FasL(sEas、sFasL),用流式细胞仪检测CD_3^+细胞Fas抗原的表达。结果 血透患者CD_3^+ Fas表达率显著高于正常对照(P<0.01);透析后Fas表达明显降低(P<0.01);但FasL表达率仍高于正常对照(P<0.05);而血浆sFasL水平比对照组明显减低(P < 0.01),透析后虽有所增加,但仍低于对照组(P<0.05)。经相关分析发现 sEas与CD_3^+Fas细胞百分率呈显著负相关;与尿素氮、肌酐呈显著正相关。结论 Fas/FasL表达率增高促使外周淋巴细胞凋亡。 Objective To investigate the effect of apoptosis of the peripheral lymphocytes induced by Fas/FasL system on the immunological function of Hemodialysis (HD) patients. Methods Plasma Ievel of sFas/sFasL and the espression of Fas antigen on peripheral blood Lymphocytes(PBL) membrane in 35 HD patients and 25 normai controls were determined by ELISA and flow cytometry respectively. Resolts The PBL of CD3+ Fas+ cells was significantly higher(P<0.01) in HD patients than the normai controls, and the PBL of CD3+Fas + cells in after HD was much lower than that in before HD(P<0.01) .The FasL content was obvi-ously higher (P<0.05) after HD patients than the normai controls.The level of sFas of HD patients were signficantly lower (P<0.01) than the normai controls.The PBL of CD3+ Fas+ cells was negatively correlated with sFas level, and CD3+ Fas+ cells was positively correlated with urea nitrogen and creatinine level in HD patients. Conclusion Fas/sFasL system aberration is involved in the pathogenesis of immune suppression in HD patients.
