破骨细胞病理生理学

摘要   破骨细胞是人体内高度专业化的细胞,其唯一功能就是分解骨.与其对立的成骨细胞之功能是制造骨质.理解成骨和破骨细胞之间的失衡机制仍然是当今人体病理生理学的一大挑战.其临床意义之重大归究于骨质疏松症的普遍性,此病是目前最主要的骨代谢性疾病.破骨细胞生理学研究的最新进展催生了几种新的骨质疏松症治疗方案.即便如此,仍然存在着许多挑战去完整地剖析成骨-破骨的体内更新周转.破骨细胞对骨质代谢进而对骨密度的综合效应依赖于两个主要变量因子:其数量和活力.其数量由细胞形成(分化)和死亡(凋亡)的相对速率来决定.而细胞内多种信号调控着其活力即破骨活力--其中心机器就是高活力耗能的质子泵.因此,本文将集中讨论关于分化,功能和凋亡这几方面的破骨细胞生理学.……
