凋亡素对R-OS-732细胞骨架的影响

摘要 目的 研究凋亡素(apoptin,VP3)对骨肉瘤OS-732多药耐株(R-OS-732)细胞骨架的影响及生长抑制作用。方法 将VP3基因克隆入载体pIRESI,获得重组质粒pIRV3;脂质体法转染R-OS-732细胞;制作细胞骨架标本。结果 实验组细胞骨架网状结构减少,浓聚,分布不均;对照组细胞骨架网状结构脉络清晰,舒展,分布也相对均匀。结论凋亡素对R-OS-732细胞生长具有抑制作用并导致细胞骨架破坏。 Objective To study the effects of apoptin on cytoskeleton constructure of the multidrug - resistant model of human osteosarcoma cell line (R-OS - 732) .Methods 361bpVP3 gene was cloned into the vector pIRESI, then the recombi-nant eukaryon expression vector Pirvp3 were transfected into R - OS - 732 cells with liposome cytoskeleton constructure of R - OS - 732 cells for the observation. Results The cytoskeleton constructure of the cells in control group was extended and clear, but the cytoskeleton constructure of the cells in the transfected by Pirvp3 group condensed or disappeared. Conclusion Apoptin can not only result in growth inhibition of R - OS - 732 but also destroy the cytoskeleton constructure of R - OS - 732 in the course of apoptin inducing apoptosis.
