子宫肿瘤与血管生成

摘要 血管生成是指在原有血管基础上以出芽方式生成毛细血管.Folkman[1]提出肿瘤的生长和转移有赖于血管生成,来自肿瘤细胞的化学信号可激活处于休眠状态的内皮细胞.通过实验,进一步提出肿瘤生长需经历两个时期:①血管前期.肿瘤生长限制在1~2 mm3,极少或不发生转移;②血管生成期.一旦新血管生成,肿瘤细胞呈指数增长,转移的几率随之增高[2].目前已证实有许多细胞因子与生长因子可促进血管生成,如血管内皮生长因子(VEGF),嗜酸性成纤维细胞生长因子(AFGF)、嗜碱性成纤维细胞生长因子(BFGF)、血小板衍生生长因子(PDGF)、转化生成因子α和β,血管调理素(angiotropin)、血管生成因子(angioge-nih)、肿瘤坏死因子-α(TNF-α)等[3],但仅VEGF是选择性内皮细胞生长因子,是由Ferrara和Gospdarowiz等分别在牛垂体滤泡星状细胞体外培养液中首先纯化出来的.其在血管生成和血管通透性改变的生理病理过程中起调控作用.其他因子虽本身无直接刺激血管生成作用,但可通过调节VEGF的表达来间接发挥其致血管生成作用[4].
