血卟啉衍生物光动力学疗法体外杀伤人喉癌细胞株Hep-2的实验研究 被引量:4

摘要 目的 :探讨使用血卟啉衍生物对喉癌细胞进行光动力学治疗的有效照光时间和有效药物使用量。方法 :使用波长 6 30nm的激光照射同血卟啉衍生物共同培养的喉癌Hep - 2细胞株 ,用MTT法评价不同的用药浓度(0 μg、5 μg、10 μg、5 0 μg、10 0 μg、2 0 0 μg ml)和不同的光照能量密度 (10J cm2 、2 0J cm2 、30J cm2 、4 0J cm2 、5 0J cm2 )细胞毒的作用。结果 :随着药物浓度的增加细胞的死亡率增加 ,但是在 5 0 μg ml以上增加不明显。随着光密度的增加细胞的死亡率增加 ,在 4 0J cm2 以后死亡率没有明显的增加。结论 :血卟啉衍生物在 6 30nm的激光照射下能够有效的在体外杀伤肿瘤细胞 。 Objective:To investigate the effective dose of haematoporphyrin derivative and the optimal irradition power density for photodynamic therapy in vitro.Methods:Hep-2 was selected as a reliable model to examine the dose(0μg\,5μg\,10μg\,50μg\,100μg\,200μg/ml)of haematoporphyrin derivative and the optimal irradition power density(10J/cm\+2\,20J/cm\+2\,30J/cm\+2\,40J/cm\+2\,50J/cm\+2)affecting the sensitivity of tumor cells to photodynamic therapy using a laser at 630 nm.MTTassays were performed to assess the cell mortality following the irradiation.Results:The results showed the dosed dependency of photodynarmic therapy and the output power-dependency.However,the increases of irradition power density(10-40J/cm\+2)and dose(5-50μg/ml)in fluence led to a significant enhancement of cytotoxicity until maximal values were reached.Conclusion:Haematoporphyrin derivative can effectively kill the Hep-2 cell in vitro.The cytotoxicitywas related with the dose and power density of the laser irradition\;
