摘要
目的建立一种快速、简便、灵敏分光光度法测定血清铜。方法在表面活性剂Tween-80存在下,用2-(3,5-二溴-2-吡啶偶氮)-5-二甲氨基酚(简称3,5-diBr-DNPAP)作显色剂直接光度法测定血清铜。结果该方法显色络色物最大吸收波长为564nm,线性范围达63.0μmol·L-1,表现摩尔吸光系数为7.13×104L·mol-1·cm-1。回收率为98.2~102.9%。批内和批间变异系数(cv)分别为2.9%与3.6%,与原子吸收分光光度法比较相关良好,Y=1.03X-0.42,r=0.9913,P>0.05,45例健康人血清铜含量为(9.0~23.8)μmol·L-1(x±2s)。结论该法血清用量少,不必去蛋白,具有操作快速、简便、结果灵敏可靠等优点,适合临床应用。
Objective To establish a quick,simple,sensitive and stable method for determination of copper in serum by UV-Spectrophotometry.Methods Using3,5-diBr-DMPAP as color developer in direct photometry for determination of serum copper in the presence of surfactant Tween-80.Results The maximum absorption of the complex compound was564nm,The linearity of test was up to63.0μmol·L -1 ,The molar absorp-tion coefficient is7.13×10 4 L·mol -1 ·cm -1 .The recovery rate ranged from98.2%to102.9%.The within-run and between-run CVs were2.9%and3.6%respectively.Com-paring this method(Y)with AAS(X),we obtained the regression equation.Y=1.03X-0.45,r=0.9913,P>0.05.Of serum copper content in45healthy humans,the reference interval(x±2s)was(9.0~23.8)μmol·L -1 (n=45).Conclusions This method requires a small quan-tity of serum only,needs not to remove protein and is rapid simlpe.sensitive,and reli-able,and it's suitable for clinical application.
