内源性NO对脑缺血再灌注后线粒体NADH-CoQ还原酶、琥珀酸脱氢酶活力的影响 被引量:1

摘要 目的:观察脑缺血和再灌注后脑细胞线粒体呼吸功能的改变,及应用NOS抑制剂LNNA对线粒体呼吸功能的保护作用.方法:测定脑缺血及再灌注不同时间线粒体呼吸链NADH-CoQ还原酶、琥珀酸脱氢酶活力,及给予LNNA后这两种酶活力的改变.结果:脑缺血30 min NADH-CoQ还原酶活力受抑制,琥珀酸脱氢酶活力无明显改变;再灌注1 h NADH-CoQ还原酶活力恢复,而再灌注24 h两种酶活力再次下降.再灌注1 h后不同时间给予LNNA两种酶活力高于再灌注对照组;再灌注时给予LNNA对两种酶活力无影响.结论:NADH-CoQ还原酶对缺血敏感,琥珀酸脱氢酶对缺血有一定耐受性;内源性NO的过量产生对线粒体具有毒性效应,于再灌注1~2 h后出现,LNNA对再灌注后线粒体呼吸功能具有保护作用.
