α-淀粉酶活性测定方法研究及测量结果不确定度评估 被引量:2

摘要 以2-氯-对硝基苯麦芽三糖苷为底物,采用初始速率法测定了α-淀粉酶的活性.通过实验考察了底物浓度、酶浓度、pH、温度、离子强度等对酶活性测定的影响.同时测定了α-淀粉酶的米氏常数,并对酶活性测定结果的不确定度进行了评估,酶活性测定的不确定度来源为酶反应过程、底物浓度测定、酶加入体积和温度的测定.
