禽流感免疫检测检验及应用

摘要 1998年以来,山西省多次发生禽流感,严重危害养禽业的健康发展.根据这些情况,我们进行了禽流感免疫检测检验的研究.对雏鸡的母源抗体和种鸡,蛋鸡免疫前后的抗体进行跟踪监测,共检测2 797份,85个鸡群其中对一个中型鸡场连续五年进行了监测,检测结果:雏鸡的母源抗体为3~5 log2.禽流感疫苗免疫鸡后30天,120天检测抗体:30天AI.H5-HI价为5~6 log2、AI-H9-HI价为5.5~6.0 log2;120天AI-H5-HI价为4.0~4.3 log2、AI-H9-HI价为4.1~4.4 log2,看来种鸡和蛋鸡按正常免疫,免疫期为120天左右.对种鸡的免疫剂量进行了跟踪检测,若种鸡140~150 日龄0.5 mL/只免疫,到260~270日龄检测免疫抗体降到临界线,在产蛋高峰期再进行一次免疫;若种鸡140~150日龄0.7~1 mL/X免疫,到305日龄左右检测免疫抗体降到临界线,可以避开产蛋高峰期再进行禽流感疫苗免疫,而且雏鸡母源抗体高、维持的时间长.根据雏鸡母源抗体的高低确定雏鸡的首免时间并制定了一个较为科学的免疫程序:雏鸡母源抗体为3~4 log2左右21~28日龄首免、140~150日龄二免,以后间隔120天左右进行三免;雏鸡母源抗体为5 log2左右35~40日龄首免、140~150日龄二免,以后间隔120天左右进行三免,这个免疫程序已在生产中应用,收到了良好的效果,保证鸡群健康生长和安全生产. Avian influenza has occurred many times in Shanxi province since 1998.It seriously influenced the healthy development of the poultry industry in this area.In terms of the situations,the study on the immunological detection of avian influenza was conducted in this work.Overall 2797 birds from 85 chicken flocks have been monitored on the maternal antibody of chicken and the changes of the antibodies of breeding birds and layers before and after immunization.The results showed that the maternal antibody of chicken is 3~5 log2.The titers of AI-H5-HI and AI-H9-HI were 5~6 log2 and 5.5~6.0 log2 detected at 30 days after vaccination,and 4.0~4.3 log2 and 4.1~4.4 log2 detected at 120 days after vaccination,respectively,which means the immune period is about 120 days for breeding hens and layers vaccinated in the normal procedure.The immune dosage of the AI vaccine for breeding layers was tracing tested.The results showed that if the bird was vaccinated with 0.5 ml at the age of 140~150 days old,the immune antibody declined to the critical line at the age of 260~270 days old,and need to be re~vaccinated during peak production.If the bird was vaccinated with 0.7~1 ml of the AI vaccine at the age of 140~150 days old,the immune antibody declined to the critical line at the age of 305 days old,and don't need to be re-vaccinated during peak production,and the maternal antibody of chicken is higher and remain longer time.In addition,the chicken's reasonable age for the first vaccination was determined and a comparatively scientific vaccination procedure was made basing on the level of the maternal antibody.When the maternal antibody of chicken is 3~4 log2,the first and second vaccinations should be done at 21~28 day-old and 140~150 day~old,respectively,and 120 days later for the third vaccination.When the maternal antibody of the chicken is about 5 log2,the ages of the first and second vaccinations is 35~40 and 140~150 day-old respectively,and the third vaccination age is still 120 days later after the second vaccination.This procedure has been already used in practice and good effects has achieved,which provide the protection for the healthy growth and safe production for the poultry industry.
