高血压血小板中蛋白激酶C底物磷酸化程度的实验研究

摘要 目的 研究血小板蛋白激酶C(PKC)在高血压中的活性 ,以揭示PKC的致病作用。方法 利用32 P NaH2 PO4 标记血小板 ,以PKC底物 40kD蛋白磷酸化作为酶活性的标志 ,对高血压组及正常对照组PKC的基础活性及反应活性进行分析比较。结果 高血压组PKC的基础活性及经凝血酶刺激后的反应活性均较正常对照组显著增高 (P <0 .0 1)。结论 提示PKC活性增高为高血压病形成过程中的重要机制之一。 Objective To study the protein kinase C(PKC)activity in platelets for the patients with essential hypertension and its role in the disease Method We labelled plateletes with 32p-NaH 2PO 4 and took the phosphorylation of platelet membrane 40 kd substrate as the mark of activated PKC We compared and analysed the basic and responsive activity of platelet PKC between essential hypertension group and control group by t-test Result The basic activity and responsive activity of platelet PKC after thrombin's stimulation in patient group were remarkably elevated than in control group( P <0.01) Conclution The unusual activated PKC played an important role in esssntial hypertension
