摘要 基质金属蛋白酶(MMPs)是一组能降解细胞外基质成分的具有重要生理和病理功能的酶。过去研究已对MMPs的生化和结构方面等了解取得了很大的进展,其中包括MMPs的活化,催化机制,底物特异性,组织金属蛋白酶抑制剂(TIMPs)的抑制机制,等,然而仍有许多重要未决问题。为此,本文对MMPs的结构,分类,信号通路,及其在心肌纤维化中的作用等进展进行探讨。 Matrix metalloproteinases(MMPs) are important components in many physiological and pathological processes because of their ability to degrade extracellular matrix(ECM) components.Considerable advancements have been made in the understanding of biochemical and structural aspects of MMPs,including their activation and catalytic mechanisms,substrate specificity,and the mechanism of inhibition by tissue inhibitors of metalloproteinases(TIMPs).Nonetheless,there are important questions that remain understanding.F...
