摘要
目的在X线辐射损伤小鼠睾丸生精上皮中Ghrelin表达变化的意义。方法采用辐射剂量1.0GyX线对成年小鼠进行全身照射,于照射后16h、2w和4w取小鼠睾丸组织制备标本,免疫组织化学ABC法检测Ghrelin的表达及变化。结果正常小鼠睾丸Ghrelin表达主要集中在睾丸间质的间质细胞,生精上皮呈阴性反应,照射后Ghrelin间质细胞为阴性,而生精上皮内出现阳性反应,阳性反应定位于精原细胞、精母细胞和早期变形精子细胞,支持细胞未见阳性反应。但随照射时间延长,Ghrelin表达逐渐减少。结论在X线辐射损伤小鼠睾丸生精上皮中Ghrelin表达的变化,表明Ghrelin及其受体GHS-R系统在小鼠睾丸辐射急性损伤及损伤后修复过程中具有一定的调节作用。
Objective ABSTRACT To investigate the expression of Ghrelin in spermatogenic epithelium of mouse testis,and to explore its significance in the process of X-ray impairment.Methods Mice received whole-body of x-ray exposure at dose level of 1.0Gy.Then,testicular tissues were prepared 16 hours,2 weeks and 4 weeks after irradiation.Imunohistochemical ABC method was used to detect the expression of Ghrelin in normal and X-ray impaired mice testis.Results The immunolocalization showed that Ghrelin was mainly localized in the cytoplasm of Leydig cell in normal mouse testis.In x-ray irradiated mouse testis,the expression of Ghrelin in Leydig cell was not observed,but the immunostianing was in spermatogonia,primary spermatocytes and elongating spermatids after mice impaired,and the immunostianing was not observed in elongating spermatids of 4 weeks grp.Conclusions The changes expression of Ghrelin showed that Ghrelin and it's receptor GHS-R system may play a regulation role in the acute impairment and reparation of X-ray irradiated mouse testis.
