摘要
目前,国内做发酵生产木糖醇研究的科技工作者很多,但大多数人所用菌种不是在国内菌种保藏机构购买,就是由国外引进。由于五碳糖不易被微生物利用,因此木糖醇生产菌种的自然筛选比较困难,至今国内还没有人对在自然界中筛选木糖醇生产菌种的方法做详细阐述。本文描述了从自然界筛选木糖醇生产菌株的一种方法,详细介绍了菌种获取和培养液检测的过程。作者分别选取玉米芯垛底土样、木糖废液、木糖离子交换液作为菌种来源载体,经富集、平板划线、摇瓶培养、培养液检测,筛得一株木糖醇高产菌种,在转速为200rpm,温度为30℃的条件下,于恒温培养箱中振荡培养72h,取培养液经滤纸过滤和0.2μm的滤膜微滤23次后,由液相色谱检测,得到的木糖醇干物质含量达79%,转化率大大高于传统的化学法。
Many domestic scientists are working on the research of xylitol fermentation processure.However,the strains used in the research are either bought from culture collection organization or from the abroad.It is difficult to screen the proper strains from thenature.So far,no report was found to illustrate natural strain screening in the production of xylitol.This article described a method of natural strain screening in detail and introduced the process as well as the analysis of culture medium.
